Chromatography methods for His-tagged protein purification
Immobilized Metal Ion Affinity Chromatography (IMAC) is the chromatography technique used for purification of His-tagged proteins. It utilizes the affinity for binding of histidine to different metal ions such as Ni2+, Co2+, Zn2+ or Cu,2+ immobilized by chelating ligands on the chromatographic resin. This method is simple to use and gives very high purity in only one step. Three different WorkBeads IMAC resins are available: WorkBeads 40 IDA, WorkBeads 40 NTA and WorkBeads NiMAC.
WorkBeads 40 IDA, WorkBeads 40 NTA can either be purchased uncharged or precharged with the different metal ions described above. WorkBeads NiMAC resin is pre-charged with very strongly bound nickel ions resulting in high resistance to reducing agents such as DTT and chelating substances such as EDTA.
GoBio Mini His-tag Screening kits are available in two column sizes, 1 mL and 5 mL, pre-packed with WorkBeads 40 NTA and WorkBeads 40 IDA. These screening kits for each chelating ligand contain ready-to-use GoBio Mini columns, charged with four different metal ions, Ni2+, Co2+, Cu2+ and Zn2+. This diversity, combined with the excellent mechanical properties and tight size distribution of WorkBeads resin, provides the perfect starting point for purification of your His-tagged protein.

WorkBeads 40 IDA
WorkBeads 40 IDA resins are based on the iminodiacetic acid (IDA) chelating groups. The resins can easily be charged, before use, with a broad spectrum of divalent or trivalent transition metal ions, including Ni2+, Co2+, Cu2+ and Zn2+, Ga3+ or Fe3+. They can then be used for the immobilized metal ion affinity chromatography (IMAC) purification of His-tagged proteins or other proteins with an affinity for metal ions, which are immobilized via a metal chelating ligand on the chromatography resin.
WorkBeads NiMAC
WorkBeads NiMAC resin is pre-charged with very strongly bound nickel ions resulting in high resistance to reducing agents such as DTT and chelating substances such as EDTA. The high stability allows purification of proteins from sources such as eukaryotic cell extracts, that normally would cause significant nickel ion stripping from the resin. This reduces the need for sample pre-treatment. The resin provides high purity and binding capacity and the possibility to use high flow rates for minimized process time.
